A promising chemical modification strategy, cationization, achieves enhanced biopolymer properties by permanently incorporating positive charges into the biopolymer backbone. Though non-toxic and abundant, carrageenan, a polysaccharide, finds frequent application within the food industry, unfortunately suffering from limited solubility in cold water. Our study involved a central composite design experiment to evaluate the parameters that had the greatest effect on cationic substitution and film solubility. Carrageenan's backbone, augmented with hydrophilic quaternary ammonium groups, promotes interactions in drug delivery systems, thus creating active surfaces. Statistical evaluation revealed that, over the specified range, only the molar ratio between the cationizing reagent and the repeating disaccharide unit of carrageenan presented a substantial effect. A 6547% degree of substitution and 403% solubility were realized by optimized parameters employing 0.086 grams of sodium hydroxide and a glycidyltrimethylammonium/disaccharide repeating unit of 683. The characterizations substantiated the effective integration of cationic groups into the carrageenan's commercial framework, thus enhancing the thermal stability of the derivative compounds.
Three types of anhydrides, differing in structure, were incorporated into agar molecules to examine how varying degrees of substitution (DS) and the anhydride structure influence physicochemical characteristics and curcumin (CUR) loading capacity in this study. Modifications to the carbon chain length and saturation of the anhydride impact the hydrophobic interactions and hydrogen bonds present in the esterified agar, thereby leading to a change in the agar's stable structure. Pectin's claimed health attributes are often linked to its fermentability in the large intestine, but in-depth research on the structural aspects of this fermentation has remained unreported. The structural variations of pectic polymers were a key focus of this study on pectin fermentation kinetics. Six commercial pectins from citrus, apple, and sugar beet varieties were chemically evaluated and subjected to in vitro fermentation with human fecal samples, monitored at different time intervals (0, 4, 24, and 48 hours). The structure of intermediate cleavage products demonstrated disparities in fermentation speed and/or rate across various pectin samples, while the sequence of pectic element fermentation exhibited similar patterns in all instances. The fermentation process first focused on the neutral side chains of rhamnogalacturonan type I, occurring between 0 and 4 hours, followed by the homogalacturonan units, fermented between 0 and 24 hours, and concluding with the rhamnogalacturonan type I backbone fermentation, which spanned from 4 to 48 hours. The potential exists for differing fermentations of various pectic structural units in different segments of the colon, impacting their nutritional value. No time-related correlation existed between the pectic subunits and the generation of diverse short-chain fatty acids, such as acetate, propionate, and butyrate, and their consequence on the microbial community. For all pectins examined, an augmentation of the bacterial genera Faecalibacterium, Lachnoclostridium, and Lachnospira was discernible.
Inter/intramolecular interactions contribute to the rigidity of the chain structures of natural polysaccharides like starch, cellulose, and sodium alginate, which contain clustered electron-rich groups, thus making them noteworthy as unconventional chromophores. Owing to the abundant hydroxyl groups and the close arrangement of low-substituted (under 5%) mannan chains, we performed an investigation into the laser-induced fluorescence of mannan-rich vegetable ivory seeds (Phytelephas macrocarpa), both in their natural form and after thermal aging. Under 532 nm (green) excitation, the untreated material emitted fluorescence light at a wavelength of 580 nm (yellow-orange). The abundant polysaccharide matrix of crystalline homomannan is demonstrably luminescent, as confirmed by lignocellulosic analyses, fluorescence microscopy, NMR, Raman, FTIR, and XRD. Thermal aging processes, conducted at temperatures of 140°C and higher, reinforced the yellow-orange fluorescence in the material, triggering its luminescent properties when activated by a near-infrared laser with a wavelength of 785 nanometers. Given the clustering-driven emission mechanism, the fluorescence of the unprocessed material is likely caused by hydroxyl clusters and the conformational rigidity found within mannan I crystals. Alternatively, thermal aging was responsible for the dehydration and oxidative breakdown of mannan chains, consequently causing the substitution of hydroxyl groups with carbonyls. The observed physicochemical adjustments possibly affected cluster organization, strengthened conformational stiffness, and therefore improved fluorescence emission.
